fa بررسی میزان شیوع باکتری بروسلا در نمونه های شیر خام و پنیر محلی گوسفندی عرضه شده در شهرستان فارسان از توابع استان چهارمحال بختیاری به روش PCR Investigation of Brucella bacteria prevalence in samples of raw milk and local sheep cheese supplied in Farsan city of Chaharmahal Bakhtiari province by PCR method بهداشت مواد غذایی Food Hygiene گزارش کوتاه Short Communication سلامت شیر و فرآورده های آن به دلیل ارزش بالای غذایی آن‌ها در تغذیه انسان از اهمیت بسزایی برخوردار می‌باشد. این میان وعده مغذی، یکی از مهم‌ترین منابعی است که توسط عوامل میکربی آلوده می‌شود. بروسلا یک کوکوباسیل گرم منفی می‌باشد که میزبان‌های آن شامل انسان، گاو، بز و گوسفند می‌باشد. بروسلا از طریق شیر و فراورده‌های لبنی انتقال یافته و در انسان سبب ایجاد تب مالت می‌شود. روش‌های سرولوژیک تشخیص بروسلا از حساسیت و دقت کافی برخوردار نبوده و دارای نتایج مثبت و منفی کاذب فراوانی می‌باشند. در این مطالعه 20 نمونه شیر خام و 20 نمونه پنیر محلی از سطح شهرستان فارسان جمع آوری گردید. DNAی نمونه‌ها به وسیله کیت DNP استخراج گردید. پس از بهینه نمودن راند اول و دوم تست،PCR  حساسیت و اختصاصیت آن مورد ارزیابی قرار گرفت. سپس تمامی نمونه‌ها به وسیله تست PCR بهینه شده مورد بررسی قرار گرفتند. از 20 نمونه شیر خام، یک نمونه (معادل 5درصد) PCR مثبت و از 20 نمونه پنیر محلی نمونه PCR مثبت مشاهده نشد. از آنجا که شیر به عنوان یک میان وعده بسیار مغذی و مفید مورد استفاده عموم قرار می گیرد، نتایج بدست آمده نشانگر این مطلب می‌باشدکه علیرغم تلاش‌های صورت گرفته جهت جلوگیری از بیماری بروسلوز، هنوز هم احتمال آلودگی شیرخام، در نواحی مختلف، به باکتری بروسلا وجود دارد.   Milk's healthand products are essential in human nutrition due to its high nutritional value. This nutritious snack is one of the most essential sources that microbial agents contaminate. Brucella is a Gram-negative coccobacillus whose hosts include humans, cattle, goats, and sheep. Brucella is transmitted through milk and dairy products and causes human malt fever. Serological methods for detecting Brucella do not have sufficient sensitivity and accuracy and have many false positive and negative results. This study collected 20 samples of raw milk and 20 samples of local cheese from Farsan City. Became NA of samples extracted by the DNP kit. After optimizing the first and second rounds of the test, PCR sensitivity and specificity were evaluated. Then all the samples were analyzed by optimized PCR test. Out of 20 raw milk samples, 1 sample (equivalent to 5%) was PCR positive; no PCR-positive sample was observed out of 20 local cheese samples.
